Linaclotide
Guanylate cyclase-C agonist for IBS-C and chronic constipation.
What it is
Linaclotide (Linzess) is a synthetic 14-amino-acid peptide guanylate cyclase-C receptor agonist FDA-approved for irritable bowel syndrome with constipation (IBS-C) and chronic idiopathic constipation. It acts locally in the GI tract with minimal systemic absorption.
Mechanism of action
Guanylate cyclase-C receptor activation increases intestinal fluid secretion and accelerates GI transit. Effects on visceral pain pathways may also contribute to symptom benefits in IBS-C.
Approved indications
- Irritable bowel syndrome with constipation
- Chronic idiopathic constipation
- Functional constipation in pediatric patients
